Tissue profiling of P450s
Surface sterilized seeds (ecotype Columbia) were plated on half-strength MS-agar medium supplemented with 1% sucrose. Seven-day old seedlings were grown at a temperature of 21oC with a 16 hr light/8 hr dark cycle and shoot and root tissues were harvested directly from these MS-agar plates. One-month-old plants were grown on these plates for one week, transferred to soil and grown for an additional 3 weeks under the same temperature and light conditions. Leaf, flower and stem tissues were harvested from these plants. The universal control RNA pool was derived from all aerial tissues of mature 1-month-old plants and spiked with a constant amount of RNA derived from roots of 7-day-old seedlings to eliminate the scaling problems associated with root-specific transcripts not being present in standard RNA pools. Tissue values are normalized relative to this universal control.
